From Jar to Jungle: A Practical Guide to Growing Tissue Culture Plantlets

When I first started my plant journey, a nursery worker casually mentioned “tissue culture plants.” I had no idea what that meant, but I knew I needed to find out.

What I discovered opened up an entirely different side of the plant world. While most people buy plants from nurseries, there’s a massive global system of micro-propagated plants grown in laboratories, produced by the thousands, and acclimated in bulk. Not long after, tissue culture plants became a full-blown hobby, spreading across auction sites, Etsy, and social media. With more accessible information, small-scale tissue culture labs began popping up everywhere; kitchens, closets, and spare rooms.

Fast forward three years: my husband now runs his own lab, I’ve gone from dabbling to working with hundreds of plants at a time, and I’ve made just about every mistake possible (some of them expensive). But I’ve also learned what works. Here are the key things I’ve found that can help you succeed with tissue culture acclimation.

Step 1: Rinse off the agar

First, that gooey substance your plant arrives in is agar. It’s packed with nutrients and hormones designed to promote rapid growth in sterile lab conditions, but outside of that environment, it becomes a contamination risk. It needs to be removed thoroughly.

I rinse my plantlets several times, then soak them in water for about five minutes. This extra step helps ensure all residue is gone and reduces the risk of bacterial or fungal issues later.

Step 2: Optional antifungal treatment

Next, I treat most plantlets with an antifungal. Not every guide includes this step, but I’ve found it helpful. Options range from diluted 3% hydrogen peroxide to iodine, Grow-Safe, or copper fungicide. I’ve had similar success with all of them, so I typically use whatever I have on hand.

The exception is carnivorous plants; they’re sensitive to additives, so I skip antifungals and extend the soaking time instead.

After treatment, I give the plantlets one final rinse in distilled water. Then I lay them on a clean paper towel to air dry while I prepare my pots and media.

Step 3: Preventing melt

This drying step is especially important for preventing “melt,” particularly in alocasias. Melt often happens when excess moisture collects on the plant inside a high-humidity environment. Letting the plant dry slightly before potting helps prevent tissue collapse. A good rule of thumb: fine, mist-level moisture is ideal; large water droplets are not.

Step 4: Heat and light matter

If you are going down this rabbit hole and you have not yet invested in a heat mat or a good plant light, now is the time to do it. Sunlight is not adequate at this stage, and it can actually create a greenhouse effect that pushes temperatures over 100 degrees and burns your tissue cultures.

A reliable and affordable option is Barrina; look for 5000K full-spectrum daylight white grow lights.


Step 5: Choosing the right media

After a lot of trial and error, here’s what I currently use:

  • Alocasias: a 60/40 mix of Fluval Stratum and perlite. This has given me nearly a 100% success rate, even with weaker plantlets.

  • Most aroids (monstera, philodendron, anthurium, raphidophora): when acclimating in bulk, I use Root Riot plugs. They offer excellent aeration, contain natural rooting hormones, and consistently produce strong results (and yes, I remove them before selling).

  • If I’m only acclimating a few plants, I use a standard aroid mix; just make sure it’s moist, not wet.

  • Begonias: 100% Fluval Stratum.

  • Carnivorous plants: use their species-specific media, but always pre-wet it so it’s evenly damp, not soggy.


Step 6: Focus on new growth

One of the most important lessons I’ve learned is not to rely on the existing leaves or roots. The original tissue culture growth isn’t adapted to real-world conditions. It’s the new roots, especially the fine, fuzzy ones, and new leaves that determine long-term success.


Step 7: Containers and humidity

Finally, containers matter more than people think.

Early on, I made the mistake of using containers that were too large or too wet. Tissue culture plants do best in snug spaces, typically no larger than 2 to 2.5 inches.

I’m cautious with self-watering containers because they can oversaturate the media, increasing the risk of rot and melt, especially with delicate or highly variegated plants. Domes can also be limiting if they’re too short.

What’s worked best for me is using small clear nursery pots placed inside clear 9–16 oz cups. This setup provides visibility, humidity control, and better overall consistency.

Now that I acclimate in bulk, I use domed 10" x 20" nursery trays with preformed cell inserts and skip individual containers altogether. My final stage of acclimation is potting plants into their long-term growing medium after removing plugs and waiting until new roots reach the edges of their containers; this is how I know they’re ready to transition successfully.
